摘要:菌株S3是从厦门石胄头近海海水中分离到的,不产生抗菌活性物质,而产黑色素的一株放线菌。在其形态、培养及生理生化特征、细胞壁组分测定等传统的分类学方法的基础上测定和分析了菌株的16S rDNA序列。结果表明,菌株S3在多种培养基上产黑色素,孢子椭圆或圆柱状,在ISP培养基上培养特征与Streptomyces Puniciscabiei基本一致,菌株S3的16S rDNA序列与S. Puniciscabiei的同源性为99.595%,鉴定其为S. Strain S3 was identified by methods of traditional and molecular biological taxonomy including determination of morphological and cultural characteristics, physiological and biochemical properties, cell wall components and 16S rDNA sequences analysis. Results showed that strain S3 produced melanin on many medium. Spores were oval or columned in ship. Cultural characteristics of strain S3 on ISP medium were in close agreement with S. Puniciscabiei. The 16S rDNA sequence of strain S3 had 99.595% similarity with that of S. Puniciscabiei. The strain S3 was identified as S. Puniciscabiei. Key words: Streptomyces, melanin, taxonmy 黑色素(Melanin)广泛存在于自然界生物体内,是一类由L-多巴、酪氨酸和半胱氨酸氧化或聚合而成的天然或合成色素[1]。它是医药、农业及化妆品行业的一种重要的生产原料。由于从动植物体内提取黑色素的生产过程繁琐、成本高,合成色素一般对人体有害,而产黑色素的微生物资源丰富,且生产工艺简单易行,产品无毒害,稳定性好。因此,研究微生物发酵产生黑色素具有很大的优势[2-4]。产黑色素的微生物有细菌、真菌及放线菌等。目前对陆境产黑色素的微生物研究较多,而对海洋中的微生物的报道较少。本研究从厦门石胄头海水中分离到一株放线菌,该菌具有产黑色素的能力,以期为开发产黑色素的海洋微生物打下基础。 1 材料与方法 1.1 材料 |
